Effects of Ginsenoside Rg3 on Adipocyte Fatty Acid Binding Protein mRNA Expression and Glycerol-3-phosphate Dehydrogenase Activity during Adipocytes Differentiation

Abstract

|
Objective: This study evaluated the effects of ginsenoside Rg3, a red ginseng component, on the adipogenesis during 3T3-L1 adipocytes differentiation.
Methods: 3T3-L1 preadipocytes were differentiated in the presence and absence of ginsenoside Rg3 for 7 days. Cytoplasmic lipid accumulation was assessed by Oil-red O stating. Adipocyte fatty acid binding protein (aP2) mRNA expression was measured by real-time PCR. Glycerol-3-phosphate dehydrogenase (GPDH) activity was quantitated by enzymatic method.
Results: Ginsenoside Rg3 at concentration of 10 ¥ìM did not show any cytotoxic effects. Ginsenoside Rg3 suppressed the lipid accumulation, a marker of adipogenesis, in a time- or dose-dependent manner. The mRNA level of aP2, which plays an important role in adipocyte differentiation, was significantly decreased by ginsenoside Rg3 treatmeant (P < 0.05). The activity of glycerol-3-phosphate dehydrogenase (GPDH), an indicator of lipid biosynthesis, was also significantly decreased in a time- or dose-dependent manner (P<0.05).
Conclusion: These results suggest that ginsenoside Rg3 inhibits adipogenesis during adipocyte differentiation, which was at least partially mediated via decreased aP2 gene expression and GPDH activity.
